ترمیم‌ فراموشی القا شده با اتانول توسط لاکتوباسیلوس رامنوسوس، پروبیوتیک بومی ایران، در رتهای نر نژاد ویستار

چکیده

پروبیوتیک‌ها مکمل‌های غذایی و دارویی تهیه شده از میکروارگانیسم‌های زنده می‌باشند که با بهبود تعادل میکروبی روده، اثرات مفیدی بر روی میزبان (انسان یا حیوان) اعمال می‌کنند. تحقیقات نشان داده که مواد غذایی از طریق اعصاب واگ می‌توانند بر روی مغز و هورمون‌های عصبی تاثیر بگذارند. از این رو این احتمال وجود دارد که پروبیوتیک‌ها بتوانند با تنظیم فعالیت‌ بخش‌هایی مختلف مغز، بر بهبود کارکرد آن موثر باشند.هدف این مطالعه بررسی تاثیر، لاکتوباسیلوس رامنوسوس، پروبیوتیک‌ بومی جدا شده از لبنیات سنتی ایران بر یادگیری احترازی غیر فعال موش صحرایی بود. در این تحقیق از 56 موش صحرایی نر نژاد ویستار با وزن تقریبی 250 تا 300 گرم استفاده شد. برای القاء فراموشی از اتانول به صورت تزریق زیرجلدی بعد از آزمون استفاده شد و همچنین گاواژ موش­ها با 1/0 میلی­لیتر بافر فسفات (PBS) حاوی ml/cfu 109 به تنهایی یا همراه با لاکتوباسیلوس رامنوسوس به مدت یک ماه صورت گرفت. آزمون رفتاری با استفاده از دستگاه یادگیری احترازی غیرفعال انجام شد. نتایج ما مشخص کرد که تزریق زیرجلدی بعد از آزمون اتانول ( mg/rat1) منجر به فراموشی می شود. این فراموشی در گروه مصرف کننده پروبیوتیک بازگردانده می­شود. یافته‌های این تحقیق نشان‌گر اثر افزایشی لاکتوباسیلوس رامنوسوس پروبیوتیک بومی جدا شده از دوغ ترخینه، در یادگیری شرطی احترازی غیرفعال در موش صحرایی نر نژاد ویستار می­باشد که تزریق زیرجلدی از اتانول را دریافت کردند. این امر نشان­دهنده اثر بهبوددهنده پروبیوتیک‌های خوراکی بر حافظه و یادگیری است.

Probiotics are food supplements and pharmaceutical substances prepared from living microorganisms that with enhancement of microbial equilibrium of intestinal microbiota, insert useful effects on the host (human/animal). Nutrients could affect the brain and neurohormones via vagus nerve. Therefore, probiotics can regulate some parts of brain activities and effect in better function. The aim of this study was examination of Lactobacillus rhamnosus, the native Iranian probiotic isolated from Tarkhineh dough (a carbonated yoghurt drink) product on the rat passive avoidance. We used 56 male Wistar rats with 250-300 gr weight range. For induced amnesia intra peritoneal pre-training injection ethanol and for one mount administrated with1×10 9 CFU/ml of with or without probiotic bacteria. We used the step-through passive avoidance to examine long-term memory in male Wistar rats. Post-training subcortical (SC) administration of ethanol (1 mg/kg) induced amnesia. This amnesia was reversed in probiotic receiving groups. L. rhamnosus isolated from Tarkhineh dough, enhanced the passive avoidance learning of male Wistar rats that received ethanol. Our data demonstrated the positive effect of this probiotic on learning and memory.
