تاثیر حفاظتی پروبیوتیک Bacillus coagulance بر ایجاد پارکینسون درموش های صحرایی نر

چکیده

بیماری پارکینسون یک اختلال نوروپاتولوژیک شایع است که به علت دژنراسیون نورون­های دوپامینرژیک بخش متراکم جسم سیاه ایجاد می­شود. هدف از مطالعه حاضر بررسی نقش پروبیوتیک باسیلوس کواگولانس، در اختلالات حرکتی ناشی از تزریق نوروتوکسین 6-هیدروکسی دوپامین در مدل پارکینسون در موش صحرایی نر می­باشد. در این مطالعه تجربی از 48 سر موش­های صحرایی نر نژاد ویستار با محدوده وزنی 200-250گرم  استفاده شد. موش­های صحرایی به صورت تصادفی به 6 گروه 8 تایی تقسیم شدند. در آزمایش اول دو گروه موش انتخاب شدند که یک گروه سالم بوده و گروه دوم توسط 6-هیدروکسی دوپامین پارکینسونی شدند. در آزمایش دوم یک گروه از موش­ها سالم بوده ، سه گروه دیگر پارکینسونی شدند که یک گروه از آنها یک ماه قبل از جراحی سالین گاواژ شد و دو گروه دیگر شیر، شیر و پروبیوتیک دریافت کردند. ایجاد مدل پارکینسونی از طریق 6-هیدروکسی دوپامین به موش­های صحرایی سبب القای رفتار خشن و اختلالات حرکتی شد. پیش تیمار موش­های نر با پروبیوتیک باسیلوس کواگولانس به مدت یک ماه انجام شد و سپس پارکینسونی شدند نتایج آزمایشات نشان داد که پروبیوتیک باسیلوس کواگولانس سبب کاهش معنی­دار اثر بیماری شد و بهبود علایم حرکتی در تست اپومورفین را به همراه داشت. براساس نتایج این مطالعه پروبیوتیک­ها باعث جلوگیری از ایجاد پارکینسون در موش­های بزرگ آزمایشگاهی می­شوند.

Protective Effect of Bacillus coagulance Probiotic in Parkinson’s Male Rats

Parkinson's disease is a common neuropathological disorder caused by the degeneration of dopaminergic neurons in substantia nigra pars compacta. This study aimed to investigate the effect of bacillus coagulance probiotic extract on the movement disorders from the injections of neurotoxin 6-hydroxy dopamine in Parkinson models in male rats. In this study, 48 male Wistar rats weighing 250-300g were used and divided into six groups, eight rats per group. In the first experiment, two groups of rats were selected in which one group is intact and other group receives 6-OHDA which was Parkinsonism. In the second experiment, 8 male rats received water and food for a month, 8 male rats received salineto a 1ml for a month, 8 male rats received Bacillus Coagulans with milkto a 1ml for a month, 8 male rats received milk to a 1 ml and then all groups receive 6-OHDA in the brain. The induction of PD by the injection of 6-hydroxydopamine results in the aggressive behavior, movement disorders. Treatment of male rats with probiotic Bacillus coagulans for one month and then induct Parkinson disease. This experiment resulted in a significant reduction in the disease which improved the motor symptoms in apomorphine test. Probiotics seems to increase the neurotransmitter dopamine in the brain and the protective effect on Parkinson's disease.
