بررسی بیان RNAهای بلند غیرکدکننده‌ی CCAT1، CCHE1، ANRIL، BANCR و PICART1 در سرطان ریه

چکیده

سرطان ریه شایع‌ترین بدخیمی با بالاترین میزان مرگ و میر در سراسر جهان است. RNAهای بلند غیر کد‌‌‌کننده (LncRNAs) نقش‌های پیچیده‌ای در شروع و پیشرفت سرطان ریه دارند و در نتیجه بر پیش‌آگهی بیماران تأثیر می‌گذارند. در پژوهش حاضر تغییرات بیان lncRNAهایCCAT1 ،CCHE1 ،ANRIL ، BANCR و PICART1 در بافت‌های توموری در مقایسه با بافت‌های طبیعی مجاور تومور بررسی شدند و پتانسیل آن‌ها به عنوان نشانگر‌های زیستی برای تشخیص و پیش‌آگهی سرطان ریه ارزیابی گردید. به منظور انجام این مطالعه مورد - شاهدی، ۳۰ نمونه‌ی بافت سرطانی ریه و ۳۰ نمونه‌ی بافت غیر سرطانی مجاور تومور جمع‌آوری گردید و پس از استخراج RNA تام و سنتز cDNA، سطح بیان lncRNAهای هدف و ژن مرجع β2M با استفاده از روش Real-time PCR SYBR Green اندازه گیری شد. آنالیز آماری داده‌های Real-time PCR، بین میزان بیان این lncRNAها در  بافت توموری و بافت نرمال اطراف تومور، تفاوت معنی داری نشان ندادند (05/0 ˃ p). همچنین ارتباط معنی داری بین سطح بیان این lncRNAها با هیچ یک از ویژگی‌های جمعیت‌شناختی و پاتولوژیک بیماران مشاهده نشد (05/0 ˃ p). بر اساس یافته‌های این مطالعه می‌توان چنین نتیجه گرفت که این lncRNAها نقشی در سرطان ریه، در بیماران ایرانی ندارند. با این حال برای تأیید نتایج این پژوهش، بررسی بیان این lncRNA ها در تعداد بیشتری نمونه لازم می‌باشد.

Lung cancer is the most common malignancy with the highest mortality worldwide. LncRNAs play complex roles in the initiation and progression of lung cancer, thereby affecting the prognosis of patients. In this study, the expression changes of CCAT1, CCHE1, ANRIL, BANCR, and PICART1 lncRNAs in tumor tissues compared to normal tissues adjacent to the tumor were investigated and their potential as biomarkers for the diagnosis and prognosis of lung cancer was evaluated. In order to conduct this case-control study, 30 samples of lung cancer tissue and 30 samples of adjacent non-cancerous tissues (ANCTs) were collected and after total RNA extraction and cDNA synthesis, the expression levels of target lncRNAs and β2M reference gene was measured using SYBR Green real-time PCR assay. The statistical analysis of Real-time PCR data showed no significant difference between the expression of these lncRNAs in tumor tissue and normal tissue adjacent to the tumor (P> 0.05). Furthermore, no significant association was observed between the expression level of these lncRNAs and any of the demographic and pathological characteristics of patients (P> 0.05). Based on the findings of this study, it can be concluded that these lncRNAs play no role in lung cancer in Iranian patients. However, to confirm our results, it is necessary to evaluate the expression of these lncRNAs in a larger number of samples.
