بررسی اثر پروتئین های نوترکیب فاکتور کشنده (LF) و فاکتور محافظت کننده (mPA) بر روی سلول‌های سرطانی انسانی

چکیده

سرطان باعث حدود 13 درصد مرگ و میر انسان­ها است و هنوز درمان مناسبی برای آن یافت نشده است. روش­های متداول درمان سرطان مانند جراحی، شیمی درمانی و رادیو درمانی دارای عوارض جانبی زیادی بوده و در بعضی موارد هیچ تاثیری بر روی درمان سرطان ندارد لذا ایجاد درمان­های مناسب و هدفمند می­تواند انقلابی در درمان سرطان ایجاد کند. از روشهای نوین می­توان ژن درمانی و استفاده از ایمونوتوکسین­ها را نام برد که می­توانند تا درصد بالایی هدفمند عمل کنند. هدف از انجام این تحقیق بررسی اثر پروتئین تغییر یافته PA که فقط به سلول‌های سرطانی متصل می­گردد همراه با فاکتور کشنده (LF) بر روی سلول‌های سرطانی می­باشد. در این راستا پروتئینهای نوترکیب mPA و LF از باسیلوس سوبتیلیس نوترکیب جدا شد و بعد از بررسی وجود باند پروتئین و غلظت آنها به ترتیب با استفاده از روش SDS-PAGE و بردفورد، اثر ترکیبی از غلظت‌های مختلف این پروتئین­ها بر روی سلول‌های سرطانی ریه، پستان، پروستات و پانکراس با روش MTT بررسی شد. نتایج پژوهش نشان داد که میزان 75 نانوگرم از پروتئین نوترکیب LF و 50 نانوگرم از پروتئین نوترکیب mPA بیشترین تاثیر در مرگ سلول‌های سرطانی داشته و باعث مرگ بیش از 98درصد انواع سلول‌های سرطانی مورد بررسی را داشت. با حصول این نتایج امید است که با تولید توکسین­های هدفمند مانند ترکیب mPA و LF بتوان یک بعد جدید به درمان سرطان اضافه شود.

Cancer causes about 13% of human deaths and has not yet been properly treated. Conventional cancer treatment methods such as surgery, chemotherapy, and radiotherapy have many side effects and in some cases have no effect on cancer treatment, so creating appropriate and targeted therapies can revolutionize cancer treatment. New methods include gene therapy and using immunotoxins, which can act up to a high percentage. This study aimed to investigate the effect of modified PA protein that binds only to cancer cells along with lethal factor (LF) on cancer cells. In this regard, recombinant mPA and LF proteins were isolated from recombinant Bacillus subtilis and after examining the presence of protein bands and their concentrations using SDS-PAGE and Bradford methods, respectively, the combined effect of different concentrations of these proteins on lung cancer cells, Breast, prostate and pancreas were examined by MTT method. The results showed that 75 ng of recombinant LF protein and 50 ng of recombinant mPA protein had the greatest effect on cancer cell death and caused the death of more than 98% of the studied cancer cells. With these results, it is hoped that a new dimension can be added to cancer treatment by producing targeted toxins such as mPA and LF.
